I've seen a few discussions about re-mapping the PTT button to launch the camera app, but I have another app that I would like to launch by pressing the PTT button since I don't intend to use the PTT Express application. Is there a way to do through through configuration or do I need an API level change in the 3rd party app or the MC40 to achieve this ?

Yes you can do this through the Settings -> Key programmer. The PTT button is mapped to Trigger-4, select the trigger-4 option, use the Shortcut tab, then select the application you'd like to launch. This configuration can be exported to be imported to other MC40s, for more information checkout the Integrator's guide for button remapping.
