On my TC21 / EC50 with Android 10 I want to persist configuration data between reinstalls of the App. I can write and read the file in /enterprise/usr. After re-install the file is still there but I get an Unauthorized Access Exception when I try to read it (Xamarin). I still can delete it. Same issue in /enterprise/usr/...-app folder
What can I do to allow the app to read files from former installations (same App ID)?

Just a thought but are you setting the file permissions correctly on the written file? Perhaps explicitly set them from your app - https://docs.microsoft.com/en-us/dotnet/api/java.io.file.setreadable?vie...
