I have not tried it myself but the sample here should exercise the SimulScan API through EMDK for Xamarin: Document Capture - Zebra Technologies Techdocs
The first thing to check would be that your device definitely supports SimulScan - there is information on finding that out here: About SimulScan - Zebra Technologies Techdocs

My mistake was that I innocently assumed the following order:
1. Get SimulScanManager instance
2. Get SimulScanReader instance
3. Set SimulScanReader config
4. Enable SimulScanReader
But this is wrong! Unfortunately the Config-Field of SimulScanReader is valid only AFTER the Reader has been enabled.
Unless this is a Bug it should be clearly documented because it is very uncommon to allow access to public properties only in a certain order...
In addition the behaviour of the SimulScanMultiTemplate object constructor is unclear to me. It seems that it fails to load templates from some directories. Sometimes it works when the Overlaod with the Android.Net.Uri parameter is used, sometimes the other one works as well, but not always...
